Enzyme Kits

The need for economic, robust, scaleable and reliable processes for the synthesis of chiral APIs and intermediates has resulted in Almac’s process chemists tuning their skills at the interface of chemistry and biology and indeed embracing biocatalysts and biocatalytic processes in organic synthesis.  This paradigm shift has resulted in biocatalysis becoming the work-horse of the chemists tool-box for chiral chemistry.

Biocatalysis is the first choice for Almac for any scale-up chemistry involving chirality.

The acceptance of the biocatalyst has evolved due to its numerous applications in chemical synthesis and its ability to accept multi functional moieties.

Almac has developed a range of hydrolases, carbonyl reductases and transaminases to provide access to a broad range of chiral acids, esters, alcohols and amines and now provides access to libraries of enzymes for customer evaluation. Almac offers a simple business model that avoids ‘IP issues’.

Development and manufacturing quantities are available on a transparent and simple £/Kg basis. Screening for customer-requested biotransformations, process optimisation and manufacture of chiral products by biocatalysis are also available.

Almac offers the following kits:

Screening kits have been built for ease of use and contain 45 commercially available hydrolases (45×500 mg), pre-prepared phosphate buffer (2×200 mL) for hydrolysis and a selection of acyl donors for selective acylation of alcohols and amines (vinyl acetate (1×5 mL), vinyl butyrate (1×5 mL), succinic anhydride (1×5 g), ethyl acetate (1×5 mL) and ethyl butyrate (1×5 mL)). There is also a detailed user guide included.

The kit contains 40 diverse CRED biocatalysts (40×50 mg), pre-prepared phosphate buffer (1×200 mL) ready for use, NAD (1×150 mg) and NADP (1×150 mg) cofactors, DMSO (1×5 mL), glucose dehydrogenase (GDH, 1x 500 mg) and glucose (1×5 g) for the recycle system. A detailed user guide is also included.

(ARED Page)- The kit contains 16 diverse ARED biocatalysts (16×50 mg), pre-prepared phosphate buffer (1×200 mL) ready for use, NAD (1×80 mg) and NADP (1×80 mg) cofactors, DMSO (10 mL), glucose dehydrogenase (GDH, 1×250 mg) and glucose (1×2.5 g) for the recycle system. A detailed user guide is also included.

(NHASE Page) – Screening kits consist of 9 NHases (9×50 mg) and 5 Nitrilases (5×50 mg), pre-prepared phosphate buffer (1×200 mL) for hydrolysis, DMSO (1×5 mL) and a user guide.

The kit contains 25 diverse pre-formulated transaminase (TAm) biocatalysts as dry powders (25×50 mg), pre-prepared phosphate buffer (150 mL). The cofactors pyridoxal-5-phosphate (PLP, 1×125 mg) and nicotinamide adenine dinucleotide (NAD, 1×250 mg) are also supplied. Glucose dehydrogenase (GDH, 1×500 mg) and glucose (1x 2.5 g) are supplied in the kit for cofactor recycling. Lactate dehydrogenase (LDH, 1x 500 mg) is supplied to drive the reaction forward by removal of the inhibitory pyruvate by-product.
