selectAZyme™ Enzyme Panels

The biocatalysis group at Almac has developed the selectAZymeTM platform consisting of a range of enzyme types including hydrolases, carbonyl reductases, transaminases, P450 monooxygenases, nitrile hydratases and nitrilases to provide access to a broad range of chiral compounds such as acids, esters, alcohols, amines and amides. These libraries of enzymes are available for customer evaluation in the form of screening panels which have been built for ease of use, containing all materials required to screen against your substrate.

Almac offers a simple business model that avoids ‘IP issues’. Development and manufacturing quantities are available on a transparent and simple £/Kg basis. In-house screening for customer-requested biotransformations, process optimisation and manufacture of chiral products by biocatalysis are also available.

Hydrolase Enzyme Screening Panel: HESK-4800
The selectAZymeTMHESK-4800 panel consists of 48 commercially available hydrolase enzymes. Hydrolases, in particular lipases, dominate the field of biocatalysis due to their commercial availability, large number available for screening, a lack of need for expensive or demanding co-factors, versatility for synthetic or hydrolytic usage, and suitability for both aqueous and solvent-based reactions. From an industrial perspective they often admirably meet the essential criteria for “industrial readiness”, such as availability, performance (turnover, selectivity, volume efficiency) and freedom from intellectual property issues. These enzymes accept a very broad range of substrates and exhibit good to excellent selectivity.

Carbonyl Reductase Enzyme Screening Panel: CESK-5000
Almac has developed the selectAZymeTM CESK-5000 panel which consists of 50 proprietary carbonyl reductases (CREDs) that show good selectivity and activity for the reduction of prochiral ketones to chiral alcohols with high optical purity. Asymmetric synthesis of chiral alcohols has the obvious advantage of 100% theoretical yield when compared to resolution technology. These enzymes accept a very broad range of substrates

Aldehyde Reductase Enzyme Screening Panel: AESK-1600
The selectAZymeTMAESK-1600 panel consists of 16 aldehyde reductases (AREDs) that are useful for the synthesis of primary alcohols by reduction of aldehydes. They can also be used to selectively reduce an aldehyde in the presence of a ketone, or in the presence of other aldehydes. These enzymes exhibit a broad substrate range.

Transaminase Enzyme Screening Panel: TESK-3600
The selectAZymeTMTESK-3600 panel consists of 36 transaminase (TAm) enzymes that can be used for the synthesis of chiral amines from prochiral ketones or to resolve racemic amines.

Nitrile Manipulator Enzyme Screening Panel: NESK-2500
The selectAZymeTMNESK-2500 panel consists of 9 nitrile hydratase (NHase) enzymes and 16 nitrilase enzymes. NHases are a class of metalloenzyme that can be used for the mild hydrolysis of nitriles to give the primary amide as the direct biotransformation product. In contrast, nitrilases hydrolyse nitriles directly to the carboxylic acid. Given the harsh conditions of acid, base or high temperatures that are often required to perform such a reaction chemically, a major benefit of nitrilases is their ability to perform nitrile hydrolysis in neutral buffer solutions at ambient temperatures. Like many other enzymes, NHases and nitrilases can effect bioresolution of racemic substrates, often with high stereoselectivity. These enzymes exhibit a broad substrate range.

The selectAZymeTM EHESK-500 panel consists of 5 epoxide hydrolase enzymes that can be used for the resolution of racemic epoxides by selective ring opening in aqueous media to provide optically active anti diols and optically active epoxides.

The selectAZymeTMPDESK-500 panel consists of 5 phenylalanine dehydrogenase enzymes that can be used for the synthesis of L-amino acids by reductive amination of 2-oxo acids.

The selectAZymeTM OxESK-1000 panel consists of 10 P450monooxygenase enzymes that can be used for selective oxidation of unactivated carbons to produce alcohols and phenols, selective epoxidation of olefins, as well as N- and O-dealkylations. These enzymes exhibit a remarkable substrate promiscuity.

Ene Reductase Enzyme Screening Panel: ERESK-1300
The selectAZymeTMERESK-1300 panel consists of 13 ene reductase (ERED) enzymes that can be used for the asymmetric reduction of electron-deficient olefins. This allows the formation of up to two new chiral centres from pro-chiral olefins. They also allow regioselective reduction in a molecule containing more than one alkene moiety.

Hyperactivated Hydrolase Enzyme Screening Panel: HAHESK-1000
Hyperactivation leads to dramatic cost reductions in any enzymatic process as the observed reaction rate enhancement is generally 20x to 100x greater than that obtained with the untreated enzyme. The selectAZymeTMHAHESK-1000 panel consists of 10 second generation hydrolase enzymes which offer superior performance when compared to standard commercially available enzymes. These enzymes can tolerate far higher substrate loadings while maintaining high E values.

Miscellaneous Enzymes
Almac’s selectAZymeTM platform also offers a variety of miscellaneous enzymes including glucose dehydrogenase (GDH) for the in situ recycling of NAD(P)H cofactor during reactions with CREDs, AREDs, transaminases, phenylalanine dehydrogenases, ene reductases and P450 monooxygenases, as well as lactate dehydrogenase (LDH) for the conversion of pyruvate to lactate during transamination reactions and dihydrofolate reductase for the conversion of imines to amines.
